Rob Green, Luke Ayling, Kyle Bartley, Liam Bridcutt, Hadi Sacko, Pablo Hernandez, Chris Wood and Kemar Roofe have all been dropped from the side.
Leeds will start the game as big favourites for the cup tie, despite playing away from Elland Road.
The Yorkshire club have been in phenomenal form recently and have won four of their last five games in the Championship.
Monk’s side have emerged as serious promotion candidates this season and are currently occupying a play-off position as they sit fifth in the league table.
Leeds have turned their attention to the FA Cup on Monday and they come up against a strong Cambridge side that are in good form.
Cambridge are currently pushing for a play-off spot in League Two and have won seven of their last eight league games, so should prove a difficult opposition for Leeds.
